Introduction
PRTN3 is a polymorphonuclear leukocyte serine protease which degrades elastin, fibronectin, laminin, vitronectin, and collagen types I, III, and IV (in vitro) and causes emphysema once managed by tracheal insufflation to hamsters.
Description
PRTN3 is a natural antigen having a molecular mass of 25kDa to 32kDa due to glycosylation isoforms. PRTN3 is isolated from human peripheral blood leukocytes.
Source
Native.
Formulation
PRTN3 is supplied in 16mM HEPES buffer pH-8.0, 400mM NaCl, and 20% glycerol.
Immunological functions
1. Binds IgG-type human auto-antibodies. Auto-antibodies to PR3 recognize conformation-dependent epitopes.
2.?Standard ELISA test (checkerboard analysis of positive/negative sera panels); immunodot test.
Applications
Western blot with rabbit anti-PR3 antisera and?mouse anti-PR3 monoclonal antibodies.
Coating concentration
0.5-1.0 μg/ml (depending on the type of ELISA plate and coating buffer). Suitable for biotinylation and iodination.
Purity
Greater than 80% in the sum of different glycosylation isoforms according to following section?as determined by SDS-PAGE and capillary electrophoresis.
Stability
Store at 4°C if entire vial will be used within 2-4 weeks. Store, frozen at -20°C for longer periods of time.?
Avoid multiple freeze-thaw cycles.
