3D Structure
ModBase 3D Structure for P15169
Specificity
This assay has high sensitivity and excellent specificity for detection of human CPN1. No significant cross-reactivity or interference between human CPN1 and analogues was observed.
Samples
Serum, plasma, tissue homogenates
Detection Range
31.25 mU/ml -2000 mU/ml.
Sensitivity
The minimum detectable dose of human CPN1 is typically less than 7.8 mU/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision
Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision
Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
Preparation and Storage
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
ISO Certification
Manufactured in an ISO 13485:2003 and EN ISO 13485:2012 Certified Laboratory.
Product Note
Select online data sheet information is drawn from bioinformatics databases, occasionally resulting in ambiguous or non-relevant product information. It is the responsibility of the customer to review, verify, and evaluate the information to make sure it matches their requirements before purchasing the kit. Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of CPN1 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS946937 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the carboxypeptidase N, polypeptide 1 (CPN1) ELISA Kit target analytes in
biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing CPN1. The ELISA analytical biochemical technique of the MBS946937 kit is based on CPN1 antibody-CPN1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CPN1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CPN1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique.
Antibody specific for CPN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CPN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CPN1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CPN1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Summary for CPN1
Carboxypeptidase N is a plasma metallo-protease that cleaves basic amino acids from the C terminal of peptides and proteins. The enzyme is important in the regulation of peptides like kinins and anaphylatoxins, and has also been known as kininase-1 and anaphylatoxin inactivator. This enzyme is a tetramer comprised of two identical regulatory subunits and two identical catalytic subunits; this gene encodes the catalytic subunit. Mutations in this gene can be associated with angioedema or chronic urticaria resulting from carboxypeptidase N deficiency. [provided by RefSeq, Jul 2008]
UniProt Comments for CPN1
CPN1: Protects the body from potent vasoactive and inflammatory peptides containing C-terminal Arg or Lys (such as kinins or anaphylatoxins) which are released into the circulation. Defects in CPN1 are the cause of carboxypeptidase N deficiency (CPND). Patients affected present some combination of angioedema or chronic urticaria, as well as hay fever or astma, and have also slightly depressed serum carboxy peptidase N, suggestive of autosomal recessive inheritance of this disorder. Belongs to the peptidase M14 family.
