Specificity
This assay has high sensitivity and excellent specificity for detection of CPN1. No significant cross-reactivity or interference between CPN1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CPN1 and all the analogues, therefore, cross reaction may still exist in some cases.

Sample Preparation

Principle of the Assay||CPN1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-CPN1 antibody and an CPN1-HRP conjugate. The assay sample and buffer are incubated together with CPN1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CPN1 concentration since CPN1 from samples and CPN1-HRP conjugate compete for the anti-CPN1 antibody . Since the number of sites is limited, as more sites are occupied by CPN1 from the sample, fewer sites are left to bind CPN1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CPN1 concentration in each sample is interpolated from this standard curve.

Carboxypeptidase N is a plasma metallo-protease that cleaves basic amino acids from the C terminal of peptides and proteins. The enzyme is important in the regulation of peptides like kinins and anaphylatoxins, and has also been known as kininase-1 and anaphylatoxin inactivator. This enzyme is a tetramer comprised of two identical regulatory subunits and two identical catalytic subunits; this gene encodes the catalytic subunit. Mutations in this gene can be associated with angioedema or chronic urticaria resulting from carboxypeptidase N deficiency. [provided by RefSeq, Jul 2008]
