The ELISA analytical biochemical technique of the MBS904353 kit is based on CUBN antibody-CUBN antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect CUBN antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, CUBN. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique.
Antibody specific for CUBN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CUBN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CUBN is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CUBN bound in the initial step. The color development is stopped and the intensity of the color is measured.

Cubilin (CUBN) acts as a receptor for intrinsic factor-vitamin B12 complexes. The role of receptor is supported by the presence of 27 CUB domains. Cubulin is located within the epithelium of intestine and kidney. Mutations in CUBN may play a role in autosomal recessive megaloblastic anemia. [provided by RefSeq, Jul 2008]

Binds to ALB, MB, Kappa and lambda-light chains, TF, hemoglobin, GC, SCGB1A1, APOA1, high density lipoprotein, and the GIF-cobalamin complex. The binding of all ligands requires calcium. Serves as important transporter in several absorptive epithelia, including intestine, renal proximal tubules and embryonic yolk sac. Interaction with LRP2 mediates its trafficking throughout vesicles and facilitates the uptake of specific ligands like GC, hemoglobin, ALB, TF and SCGB1A1.
