Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate PIGW in samples. An antibody specific for PIGW has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPIGW present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PIGW is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PIGW bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is an inositol acyltransferase that acylates the inositol ring of phosphatidylinositol. This occurs in the endoplasmic reticulum and is a step in the biosynthesis of glycosylphosphatidylinositol (GPI), which anchors many cell surface proteins to the membrane. Defects in this gene are a cause of the age-dependent epileptic encephalopathy West syndrome as well as a syndrome exhibiting hyperphosphatasia and cognitive disability (HPMRS5).
