CBLC belongs to a family of ubiquitin ligases that are negative regulators of tyrosine kinase signaling downstream of receptor tyrosine kinase and lymphocyte antigen receptors. Monoubiquitin tag promotes sorting of activated receptor and associated proteins into internal vesicles facilitating their lysosomal degradation, while polyubiquitin tag promotes proteasomal degradation. CBLC is expressed in epithelial cells.

Immunoprecipitation and Western blot of Affinity Purified anti-Cbl-c antibody shows detection of a pre-dominant band at ~52 kD corresponding to Cbl-c (arrowhead) in transfected cell lysates (left panel). Lysates are from Hek 293T cells transfected with empty vector or with Cbl-c. The predicted size of Cbl-c is 52 kD. Size markers in kD are shown to the left of the panel. This gene encodes a member of the Cbl family of E3 ubiquitin ligases. Cbl proteins play important roles in cell signaling through the ubiquitination and subsequent downregulation of tyrosine kinases. Expression of this gene may be restricted to epithelial cells, and alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene.
