The ELISA analytical biochemical technique of the MBS700414 kit is based on VLDLR antibody-VLDLR antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect VLDLR antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, VLDLR. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Introduction: The very low density lipoprotein receptor (VLDL-r) is a cell-surface molecule specialized for the internalization of multiple diverse ligands. This receptor has been suggested to be important for the metabolism of apolipoprotein E (apoE)-containing lipoprotein particles, such as very-low-density-lipoprotein (VLDL), beta-migrating VLDL and intermediate-density lipoprotein. Its structure is similar to the low-density lipoprotein receptor (LDL-r), although the two have substantially different systemic distributions and regulatory pathways. It is also one of the receptors of reelin, an extracellular matrix protein which regulates the processes of neuronal migration and synaptic plasticity.
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to VLDLR. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for VLDLR and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain VLDLR, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of VLDLR in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Mutations in this gene cause VLDLR-associated cerebellar hypoplasia. Alternative splicing generates multiple transcript variants encoding distinct isoforms for this gene.
