The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing LRP2. The ELISA analytical biochemical technique of the MBS166977 kit is based on LRP2 antibody-LRP2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect LRP2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, LRP2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human LRP2 antibody. LRP2 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human LRP2 Antibody is added and binds to LRP2 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated LRP2 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human LRP2. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

The protein encoded by this gene, low density lipoprotein-related protein 2 (LRP2) or megalin, is a multi-ligand endocytic receptor that is expressed in many different tissues but primarily in absorptive epithilial tissues such as the kidney. This glycoprotein has a large amino-terminal extracellular domain, a single transmembrane domain, and a short carboxy-terminal cytoplasmic tail. The extracellular ligand-binding-domains bind diverse macromolecules including albumin, apolipoproteins B and E, and lipoprotein lipase. The LRP2 protein is critical for the reuptake of numerous ligands, including lipoproteins, sterols, vitamin-binding proteins, and hormones. This protein also has a role in cell-signaling; extracellular ligands include parathyroid horomones and the morphogen sonic hedgehog while cytosolic ligands include MAP kinase scaffold proteins and JNK interacting proteins. Recycling of this membrane receptor is regulated by phosphorylation of its cytoplasmic domain.
