We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. The ELISA analytical biochemical technique of the MBS735556 kit is based on Activin-A antibody-Activin-A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Activin-A antigen targets in samples. Principle of the Assay||ACTA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ACTA antibody and an ACTA-HRP conjugate. The assay sample and buffer are incubated together with ACTA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACTA concentration since ACTA from samples and ACTA-HRP conjugate compete for the anti-ACTA antibody . Since the number of sites is limited, as more sites are occupied by ACTA from the sample, fewer sites are left to bind ACTA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACTA concentration in each sample is interpolated from this standard curve.

The inhibin beta A subunit joins the alpha subunit to form a pituitary FSH secretion inhibitor. Inhibin has been shown to regulate gonadal stromal cell proliferation negatively and to have tumor-suppressor activity. In addition, serum levels of inhibin have been shown to reflect the size of granulosa-cell tumors and can therefore be used as a marker for primary as well as recurrent disease. Because expression in gonadal and various extragonadal tissues may vary severalfold in a tissue-specific fashion, it is proposed that inhibin may be both a growth/differentiation factor and a hormone. Furthermore, the beta A subunit forms a homodimer, activin A, and also joins with a beta B subunit to form a heterodimer, activin AB, both of which stimulate FSH secretion.
