Principle of the Assay||ACTA ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ACTA antibody and an ACTA-HRP conjugate. The assay sample and buffer are incubated together with ACTA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACTA concentration since ACTA from samples and ACTA-HRP conjugate compete for the anti-ACTA antibody . Since the number of sites is limited, as more sites are occupied by ACTA from the sample, fewer sites are left to bind ACTA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards.                             The inhibin beta A subunit joins the alpha subunit to form a pituitary FSH secretion inhibitor. Inhibin has been shown to regulate gonadal stromal cell proliferation negatively and to have tumor-suppressor activity. In addition, serum levels of inhibin have been shown to reflect the size of granulosa-cell tumors and can therefore be used as a marker for primary as well as recurrent disease. Because expression in gonadal and various extragonadal tissues may vary severalfold in a tissue-specific fashion, it is proposed that inhibin may be both a growth/differentiation factor and a hormone. Furthermore, the beta A subunit forms a homodimer, activin A, and also joins with a beta B subunit to form a heterodimer, activin AB, both of which stimulate FSH secretion. Finally, it has been shown that the beta A subunit mRNA is identical to the erythroid differentiation factor subunit mRNA and that only one gene for this mRNA exists in the human genome.
