Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team

The ELISA analytical biochemical technique of the MBS746396 kit is based on P2Y12 antibody-P2Y12 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect P2Y12 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, P2Y12. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Principle of the Assay: P2Y12 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-P2Y12 antibody and an P2Y12-HRP conjugate. The assay sample and buffer are incubated together with P2Y12-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the P2Y12 concentration since P2Y12 from samples and P2Y12-HRP conjugate compete for the anti-P2Y12 antibody . Since the number of sites is limited, as more sites are occupied by P2Y12 from the sample, fewer sites are left to bind P2Y12-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The P2Y12 concentration in each sample is interpolated from this standard curve.

The product of this gene belongs to the family of G-protein coupled receptors. This family has several receptor subtypes with different pharmacological selectivity, which overlaps in some cases, for various adenosine and uridine nucleotides. This receptor is involved in platelet aggregation, and is a potential target for the treatment of thromboembolisms and other clotting disorders. Mutations in this gene are implicated in bleeding disorder, platelet type 8 (BDPLT8). Alternative splicing results in multiple transcript variants of this gene. [provided by RefSeq, Jul 2013]
