Specificity
This assay has high sensitivity and excellent specificity for detection of FGF-23. No significant cross-reactivity or interference between FGF-23 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between FGF-23 and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate
Detection Range
50-1000pg/mL
Preparation and Storage
Store all reagents at 2-8 degree C.
Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team
Product Note
Select online data sheet information is drawn from bioinformatics databases, occasionally resulting in ambiguous or non-relevant product information. It is the responsibility of the customer to review, verify, and evaluate the information to make sure it matches their requirements before purchasing the kit. Our ELISA Kit assays are dynamic research tools and sometimes they may be updated and improved. If the format of this assay is important to you then please request the current manual or contact our technical support team with a presales inquiry before placing an order. We will confirm the current details of the assay. We cannot guarantee the sample manual posted online is the most current manual.
Other Notes
Small volumes of FGF-23 elisa kit vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

MBS741661 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Fibroblast growth factor 23 (FGF-23) ELISA Kit target analytes in
biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing FGF-23. The ELISA analytical biochemical technique of the MBS741661 kit is based on FGF-23 antibody-FGF-23 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect FGF-23 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, FGF-23. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This FGF-23 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine FGF-23. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
Principle of the Assay: FGF-23 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FGF-23 antibody and an FGF-23-HRP conjugate. The assay sample and buffer are incubated together with FGF-23-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FGF-23 concentration since FGF-23 from samples and FGF-23-HRP conjugate compete for the anti-FGF-23 antibody . Since the number of sites is limited, as more sites are occupied by FGF-23 from the sample, fewer sites are left to bind FGF-23-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FGF-23 concentration in each sample is interpolated from this standard curve.

NCBI Summary for FGF-23
This gene encodes a member of the fibroblast growth factor family of proteins, which possess broad mitogenic and cell survival activities and are involved in a variety of biological processes. The product of this gene regulates phosphate homeostasis and transport in the kidney. The full-length, functional protein may be deactivated via cleavage into N-terminal and C-terminal chains. Mutation of this cleavage site causes autosomal dominant hypophosphatemic rickets (ADHR). Mutations in this gene are also associated with hyperphosphatemic familial tumoral calcinosis (HFTC). UniProt Comments for FGF-23
FGF23: Regulator of phosphate homeostasis. Inhibits renal tubular phosphate transport by reducing SLC34A1 levels. Upregulates EGR1 expression in the presence of KL. Acts directly on the parathyroid to decrease PTH secretion. Regulator of vitamin-D metabolism. Negatively regulates osteoblast differentiation and matrix mineralization. Defects in FGF23 are the cause of autosomal dominant hypophosphataemic rickets (ADHR). ADHR is characterized by low serum phosphorus concentrations, rickets, osteomalacia, leg deformities, short stature, bone pain and dental abscesses. Defects in FGF23 are a cause of hyperphosphatemic familial tumoral calcinosis (HFTC). HFTC is a severe autosomal recessive metabolic disorder that manifests with hyperphosphatemia and massive calcium deposits in the skin and subcutaneous tissues. Belongs to the heparin-binding growth factors family.
