Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate CHAF1A in samples. An antibody specific for CHAF1A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCHAF1A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CHAF1A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CHAF1A bound in the initial step. The color development is stopped and the intensity of the color is measured.

Core component of the CAF-1 complex, a complex thought to mediate chromatin assembly in DNA replication and DNA repair. Assembles histone octamers onto replicating DNA in vitro. CAF-1 performs the first step of the nucleosome assembly process, bringing newly synthesized histones H3 and H4 to replicating DNA; histones H2A/H2B can bind to this chromatin precursor subsequent to DNA replication to complete the histone octamer. CHAF1A binds to histones H3 and H4. It may play a role in heterochromatin maintenance in proliferating cells by bringing newly synthesized cbx proteins to heterochromatic DNA replication foci.
