MBS2000201 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the SH3 Domain Binding Protein 2 (SH3BP2) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing SH3BP2. The ELISA analytical biochemical technique of the MBS2000201 kit is based on SH3BP2 antibody-SH3BP2 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect SH3BP2 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, SH3BP2. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to SH3 Domain Binding Protein 2 (SH3BP2). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to SH3 Domain Binding Protein 2 (SH3BP2). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain SH3 Domain Binding Protein 2 (SH3BP2), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of SH3 Domain Binding Protein 2 (SH3BP2) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene has an N-terminal pleckstrin homology (PH) domain, an SH3-binding proline-rich region, and a C-terminal SH2 domain. The protein binds to the SH3 domains of several proteins including the ABL1 and SYK protein tyrosine kinases , and functions as a cytoplasmic adaptor protein to positively regulate transcriptional activity in T, natural killer (NK), and basophilic cells. Mutations in this gene result in cherubism. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Mar 2009]

SH3BP2: an adaptor protein that binds differentially to the SH3 domains of certain proteins. Links the hemopoietic tyrosine kinase Fes to the cytoplasmic membrane in a phosphorylation dependent manner. Regulates FC-epsilon RI -mediated degranulation in rat basophilic leukemia RBL-2H3 cells. Expressed in a variety of tissues including lung, liver, skeletal muscle, kidney and pancreas. Defects in 3BP2 are the cause of cherubism, an autosomal dominant inherited syndrome. Two differentially spliced isoforms have been described.
