The ELISA analytical biochemical technique of the MBS161178 kit is based on GM2A antibody-GM2A antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect GM2A antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, GM2A. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: This sandwich kit is for the accurate quantitative detection of human GM2 Ganglioside Activator (also known as GM2A) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human GM2A antibody. GM2A present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human GM2A Antibody is added and binds to GM2A in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated GM2A antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human GM2A. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

This gene encodes a small glycolipid transport protein which acts as a substrate specific co-factor for the lysosomal enzyme beta-hexosaminidase A. Beta-hexosaminidase A, together with GM2 ganglioside activator, catalyzes the degradation of the ganglioside GM2, and other molecules containing terminal N-acetyl hexosamines. It extracts single GM2 molecules from membranes and presents them in soluble form to beta-hexosaminidase A for cleavage of N-acetyl-D-galactosamine and conversion to GM3.
