Figure: Detection of endogenous MyD88 in mouse EL-4 and human HEK 293T cells.Method: Cell extracts from EL-4 (1) or 293T cells (2) were resolved by SDS-PAGE under reducing conditions, transferred to nitrocellulose and incubated with the antiserum to MyD88 at a dilution of 1:8000. Proteins were visualized using a peroxidase-conjugated antibody to rabbit IgG and a chemiluminescence detection system.

This gene encodes a cytosolic adapter protein that plays a central role in the innate and adaptive immune response. This protein functions as an essential signal transducer in the interleukin-1 and Toll-like receptor signaling pathways. These pathways regulate that activation of numerous proinflammatory genes. The encoded protein consists of an N-terminal death domain and a C-terminal Toll-interleukin1 receptor domain. Patients with defects in this gene have an increased susceptibility to pyogenic bacterial infections. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Feb 2010]

1. Authors demonstrated that MyD88 lies upstream of BLT2 in LPS-potentiated invasiveness and that this 'MyD88-BLT2' cascade mediates activation of NF-kappaB and the synthesis of IL-6 and IL-8 critical for the invasiveness and aggression of breast cancer cells.
