Specificity
Specific for the ~85kD Tlk1 (Tousled-like kinase 1) protein. It does not crossreact with the related Tlk2 protein. Antibody specificity was confirmed by Western blot with epitope-tagged Tlk1 and Tlk2 proteins, Western blot and immunoprecipitation with HeLa cell lysates and by immunoprecipitation with in vitro transcribed and translated Tlk1 and Tlk2. A nuclear localization of this protein is observed by immunohistochemistry with HeLa cells. Species Crossreactivity: human.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Small volumes of anti-TLK1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. TLK1: a protein kinase of the TLK family. Rapidly and transiently inhibited by phosphorylation following the generation of DNA double-stranded breaks during S-phase. This is cell cycle checkpoint and ATM-pathway dependent and appears to regulate processes involved in chromatin assembly. Isoform 3 phosphorylates and enhances the stability of the t-SNARE SNAP23, augmenting its assembly with syntaxin. Cell-cycle regulated, maximal activity in S-phase. Inactivated by phosphorylation, possibly by CHK1. Three splice variant isoforms have been described.
