MBS2880350 is a ready-to-use microwell, strip plate ELISA (enzyme-linked immunosorbent assay) Kit for analyzing the presence of the Wilms tumor protein (Wt1) ELISA Kit target analytes in biological samples. The concentration gradients of the kit standards or positive controls render a theoretical kit detection range in biological research samples containing Wt1. The ELISA analytical biochemical technique of the MBS2880350 kit is based on Wt1 antibody-Wt1 antigen interactions (immunosorbency) and an HRP colorimetric detection system to detect Wt1 antigen targets in samples. The ELISA Kit is designed to detect native, not recombinant, Wt1. Appropriate sample types may include undiluted body fluids and/or tissue homogenates, secretions. Quality control assays assessing reproducibility identified the intra-assay CV (%) and inter-assay CV(%).

Intended Uses: For the quantitative detection of Rat Wilms tumor protein (WT1) concentration in serum, plasma and other biological fluids.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate WT1 in Rat serum, plasma. An antibody specific for WT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any WT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for WT1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of WT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

It plays an essential role in the normal development of the urogenital system, and the human gene is mutated in a small subset of patients with Wilm's tumors. Authors of PMID:7926762 provide evidence that Wt1 mRNA undergoes RNA editing in human and rat, and that this process is tissue-restricted and developmentally regulated. Regulates the expression of numerous target genes, including EPO. Plays an essential role for development of the urogenital system. It has a tumor suppressor as well as an oncogenic role in tumor formation. Function may be isoform-specific: isoforms lacking the KTS motif may act as transcription factors. Isoforms containing the KTS motif may bind mRNA and play a role in mRNA metabolism or splicing. Isoform 1 has lower affinity for DNA, and can bind RNA.
