Immunogen
Synthetic peptide corresponding to amino acids surrounding phosphorylated Ser329 of human FKHR (KLH conjugated). The immunizing sequence is identical in mouse and rat.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Other Notes
Small volumes of anti-FOXO1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

Application Notes for anti-FOXO1 antibody
Suitable for use in Western Blot.
Dilution: Western Blot: 1:3000-1:10,000. Detected 400ng of F4523-15 phosphorylated with 0.5ug A1125-06. 400ng of F4523-15 and FKHR fusion protein incubated with 500ng Akt1/PKBa were resolved by electrophoresis, transferred to nitrocellulose and probed with F4523-21(1:3000). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
Note: This antibody is unlikely to detect endogenous phospho-FKHR in cell lysates via straight Westrern Blot analysis.

NCBI Summary for FOXO1
This gene belongs to the forkhead family of transcription factors which are characterized by a distinct forkhead domain. The specific function of this gene has not yet been determined; however, it may play a role in myogenic growth and differentiation. Translocation of this gene with PAX3 has been associated with alveolar rhabdomyosarcoma. [provided by RefSeq, Jul 2008]
UniProt Comments for FOXO1
FOXO1A: a transcription factor of the forkhead family. A central regulator of metabolism in several cell types. May play an important role in myogenic growth and differentiation, and in the regulation of lipolysis in adipocytes by controlling the expression of adipose triglyceride lipase (ATGL), the rate-limiting lipolytic enzyme. Translocation of this gene with PAX3 has been associated with alveolar rhabdomyosarcoma. Contains 1 fork-head domain.

Research Articles on FOXO1
1. Transcription factor FoxO1 transgene is a key regulator that reprograms and steers the differentiation of effector cells to functionally competent memory cells, thereby regulating the quality of CD8 T cell memory to intracellular pathogens.
