This assay has high sensitivity and excellent specificity for detection of GNRHR-Ab. No significant cross-reactivity or interference between GNRHR-Ab and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between GNRHR-Ab and all the analogues, therefore, cross reaction may still exist in some cases.

Sample Preparation
We suggest pre-experimenting with neat (undiluted) samples, 1:2 or 1:4 dilutions. Please avoid diluting your samples more than 1:10 as it would exceed the dilution limit set for this kit. If the expected concentration of the target is beyond the detection range of the kit, please contact our technical support team

Principle of the assay: GNRHR-Ab ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GNRHR-Ab antibody and an GNRHR-Ab-HRP conjugate. The assay sample and buffer are incubated together with GNRHR-Ab-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GNRHR-Ab concentration since GNRHR-Ab from samples and GNRHR-Ab-HRP conjugate compete for the anti-GNRHR-Ab antibody . Since the number of sites is limited, as more sites are occupied by GNRHR-Ab from the sample, fewer sites are left to bind GNRHR-Ab-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GNRHR-Ab concentration in each sample is interpolated from this standard curve.

This gene encodes the receptor for type 1 gonadotropin-releasing hormone. This receptor is a member of the seven-transmembrane, G-protein coupled receptor (GPCR) family. It is expressed on the surface of pituitary gonadotrope cells as well as lymphocytes, breast, ovary, and prostate. Following binding of gonadotropin-releasing hormone, the receptor associates with G-proteins that activate a phosphatidylinositol-calcium second messenger system. Activation of the receptor ultimately causes the release of gonadotropic luteinizing hormone (LH) and follicle stimulating hormone (FSH). Defects in this gene are a cause of hypogonadotropic hypogonadism (HH). Alternative splicing results in multiple transcript variants encoding different isoforms. More than 18 transcription initiation sites in the 5' region and multiple polyA signals in the 3' region have been identified for this gene. [provided by RefSeq, Jul 2008]

This receptor mediates its action by association with G-proteins that activate a phosphatidylinositol- calcium second messenger system. Isoform 2 may act as an inhibitor of GnRH-R signaling. Defects in GNRHR are a cause of idiopathic hypogonadotropic hypogonadism (IHH).
