Specificity
Recognizes phosphorylated human IGF-1R, Mr ~95kD. Also corresponds to tyrosine 1185 in human insulin receptor preprotein, containing a 27 amino acid signal peptide. Crossreacts with phosphorylated insulin and PDGF receptors which contain the I Y E motif. Slight crossreactivity detected with EGFR in A431 cells expressing very high levels of EGFR. Species Crossreactivity: mouse. Broad species crossreactivity predicted based on sequence homology. The immunizing sequence is conserved from Drosophila to mammals.

Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Small volumes of anti-IRS4 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Application Notes for anti-IRS4 antibody
Suitable for use in Western Blot and Immunoprecipitation.
Dilution: Western Blot: 0.5-2ug/ml detects phosphorylated IGF-1R in RIPA lysates from stable, transfected 3T3 cells expressing the human IGF-1 receptor treated with IGF-1 and in IGFR-containing immune complexes from IGF-1 treated, but not untreated cell lysates. Lysates from untreated and IGF-1 treated 3T3 cells expressing human IGF-1 receptor were resolved by electrophoresis, transferred to nitrocellulose and probed with I7661-18A (2ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
Immunoprecipitation: 5ug immunoprecipitates phosphorylated IGF-1 receptors from RIPA lysate from 3T3 cells treated with IGF-1.
